Partec vs Calibur.... (fwd)

I am currently caring for a patient diagnosed elsewhere with CLL
clinically 5 years ago. His counts were stable for 1 1/2 yrs and then he
was lost to followup. He was clinically stable until presenting two weeks
ago with a febrile illness,
lymphadenopathy and the following lab work:

WBC 540K
Hct 36
Plt 81
No evidence of DIC

On peripheral smear he has both small lymphocytes (predominant), larger
lymphoid appearing cells, and a
significant number (5%) of monocytoid appearing cells with abundant
somewhat granulated cytoplasm, folded nuclei, eccentric nuclei, rarely
nucleolated. There are relatively rarer prolymphs. the bone marrow is
similarly involved.

The lymphoid cells mark as monoclonal B-cells CD5,19,22,23,25,DR+ lambda
light chain positive (strong, not dim)

the nonlymphoid, monocytoid cells stain CD4,7,10,33,13,14,15,DR+ and are
CD34 negative.

Could anyone comment on this monocytoid population and its implications
for a coexistent myelodysplastic syndrome vs. M5 AML?

Thank you.

Subject: Partec vs Calibur....

Hello everybody,

I know that this has been discussed a bit, but I would like to know from
some Partec users their experience with the machines... especially
regarding:

- immunophenotyping
- bacterial analysis
- mycoplasms
- software
- compensation
- sensitivity
- sorting option

I know the FACScalibur very well (and I am very happy with it..) but I have
never really played with a Partec machine and have a friend who wanted to
know the differences between a Calibur 3 Colors and a Partec/Dako
PAS/Galaxy 3 Colors (one 488 laser). I have read the specs on the web, but
I guess real world experience is worth much more than commercial
arguments... therefore feel free to send me your comments, but it has to be
fast, because they have to make a decision by the end of the week...

thanks very much,

Matthias
