In human cells, we utilize a CD3/CD56 (BDIS)/V-alpha 24 (Immunotech) combination that gives good separation of the NKT cells from other lymphocytes. In mouse, it appears that TCR+NK1.1+ cells are your best bet. But if your mouse happens to be NK1.1-, then I have seen TCR+CD4-CD8- cells referred to as NKT cells (read Alan Baxter's papers). There have been papers published citing a V alpha 14 antibody for the mouse, but in our hands this performed unacceptably. Bendelac reported that this antibody does not bind to the V alpha 14 chain on NK-T cells, and the investigators that created the clone claim it must be run on an EPICS machine, and no other will work properly.
