ROI measurement with DHRh123

Does anyone have experience measuring reactive oxygen intermediates(ROI) by flow
with dihydrorhodamine 123?  I'm helping a user of our Core facility with this
assay.  We are finding that untreated control L929 cells have over a log shift
in green fluorescence (oxidation of DHR123 to Rh123) as compared to
autofluorescence in unloaded cells, with only modest increase in green
fluorescence in cells treated with compounds known to induce ROI production.
The user is surprised at the high basal level of ROI in these cells.
