viability of cardiac myocytes

I have aquestion concerning viability measurement of cardiac myocytes (mouse).
In our test model we need to know the cell viablity rate under several
defined conditions.
We tried propidium iodide unsuccessfully as there was no staining that
could be correlated to necrotic myocytes. Can this be due to the high
autofluorescence of cardiac myocytes?
Another fact seems to be that myocytes are susceptible to mechanical stress
occuring during analysis.

Can anyone help with these problems?
