concerning the discussion about TO-PRO-3 I just want to mention, that I got a 
paper accepted about the use of TO-PRO-3 instead HOECHST dye for the 
detection of BrdUrd together with PI for total DNA measurement. It is about 
as good as the old BrdUrd Hoechst quenching technique, but does not require a 
UV laser. May be that is of interest for you all.
