We use the following combination of antibodies in our routine panel for detecting normal and abnormal neutrophils. CD15/CD33/CD34/CD45 , CD11b/CD16/CD34/CD45, CD36/CD64/CD34/CD45. As granulocytic precursors mature you will see a gradual increase of the mature markers like CD64 and CD16 and loss of the immature markers CD34 and CD33 . This is the route of maturation from the most immature to mature ones CD34-CD33-CD15-CD11b-CD64-CD16 . Segs also have CD10, So neutrophils with CD10 and CD16 are likely your most mature cells(Segs). We devised these combinations not only to pick up normally maturing neutrophils but also to detect small populations of aberrant or neoplastic myeloid/granulocytic cells. Hope this helps.
