Just my two cents on doublet discrimination. Doublet discrimination by pulse height vs pulse width does work but not perfectly. A doublet is a non spherical particle. Effective discrimination will be dependent on the orientation of the doublet as it passes through the laser. Discrimination is also highly dependent on spot size of the beam, hence will vary with instrument configuration. I will be happy to discuss some ancient unpublished data on this subject with anyone interested. Please Email me directly. Tom Sawyer MCO Flow Cytometry Laboratory.
