Hello, I am still quite a rookie in the field of flow. I have recently come across a problem which many of you might be familiar with, the question of how to interpret ambiguous data. I have been trying to identify homozygous phenotype fas ligand negative (CD95-) cells from mouse pups that come from mixed phenotype litters. In my tests and antibody titration, it was easy to differentiate homozygous positive (CD95+ ; normal) from homozygous negative (CD95-;mutant). However, in my assays, I have found that the results are more difficult to interpret. I expected that my heterozygotes would simply have half the level of expression, but that may not be correct. Also, I'm not sure which is the best parameter by which to compare these results. It could just be a case of bad data, but I am trying to salvage as much as possible. Any help and insight would be deeply appreciated. thanks
