Hi Bob, The method is hardly top secret. Basically you open the file containing the calibration beads you used (i.e. Quantabrite or the other suppliers that make the appropriate standards). You then draw a marker around each peak and enter the number of binding sites corresponding to each peak (given by the manufacturer for each different lot of beads). FCS Express will then calculate the appropriate regression and automatically apply this regression whenever this parameter is used. Thus, statistics and the axes of plots, can display fluorescence values or binding sites, whichever you wish. Of course, you can always enter an appropriate F:P ratio if it differs from 1:1 and save and load different formulas. I found it saved a lot of time between jumping back and forth from the spreadsheet.
