Hello Mireia, I don't think that you are the only one to have come across this problem which does seem to get largely ignored. I suspect that most people are interested in the early apoptotic population (Annexin+/PI-) and just lump together all the PI+ cells together into a "late apoptotic/necrotic" population. This is fine I think until you are called upon to define the Annexin-/PI+ population. At this stage you should bear in mind one of the Flow Commandments: "Stray ye not too far from a fluorescence microscope". You could look at your cell prep (or even sort the relevant populations onto a slide) and see what is what. Hazarding a guess, these cells could be very late dead cells that have lost a large part of their membranes. Derek On Tue, 26 Jan 1999, Mireia Dalmau (S.
