Post-Sort Cell Death

Hello Fellow Sorters,

I know this topic has previously been discussed, but we have not had
post-sort cell death until recently and previous suggestions do not really
apply here.

Three months ago we moved our Vantage into our new facility. The only
difference between the old set-up and the current one is the fact that we
are using a chiller to cool the laser. Nothing else. Same 3% H2O2
sterilization procedure, same sheath fluid (PBS, Ca++, Mg++, pH 7.2) but
different water source, and no temperature problems. Sorts are going very
well but cells put into culture experience apoptosis within two days.

We've done the control experiments (unsorted, unstained cells; unsorted,
stained cells; and sorted, stained cells). The only ones that died were the
sorted cells. So... any ideas, suggestions, and prayers would be greatly
appreciated.

Thanks,

Elizabeth T. Kraus
