Hello Slava I have recently done intracellular IFNg in T and NK cells (in mice). I incubated them cell with and without PMA/Iono (100/100ng/ml) and IFNa/b (I'm studying NK cells and IFNa/b). The incubation was for four hours (my IFNa/b incubation) in the presence of monensin. The separation was never very good, but approx. 30-80 percent of NK and 20-70 of CD8 made IFNg. Hope that helps.
