We are about to upgrade FlowJo to add a sophisticated platform for specialized analyses of kinetics data. Using this platform, you can easily quantitate values including maximum response, response time, response rates (i.e., slopes), percent of cells responding, and other statistics on time-dependent data. In addition, the platform has been integrated into the FlowJo environment; thus, it supports the intelligent batch analyses operations (i.e., analyzing groups of samples, drag-and-drop analysis, etc.). We are looking for a few sites to help us with beta-testing, as well as providing ideas for further development of this platform. We are looking for people who are currently doing kinetics experiments and have access to PowerPC Macintoshes. There are a couple of examples that show analysis, in gated subsets of T cells, of calcium flux after stimulation measured either by Indo-1 fluorescence ratios, or by the percentage of responding cells.
