I have a client who has been experimenting with Clontech's ApoAlert Annexin V Kit. It uses Annexin-FITC and PI in tandem. They specify that we read the PI on the channel "reserved for PE". I have called them three times to find out why they specify this setup. No one there has been able to answer this question. Any suggestions? Additionally, the way we interpret the User Manual (which is all I have in the way of help) we should see the dead cells, early and late apoptotic cells in individual populations in the doulbe parameter histograms, ie: early apoptotic-green only, dead cells-red only, and mid apoptotic green and red. We however are not seeing this. We get a green only population and then two relatively difficult to distinguish populations in the double positive quadrant, making it impossible to determine the difference between dead and late apoptotic cells. Is anyone using this kit and having better luck, or having better luck with customer relations department. > Thanks in advance. > Joan Kalnitsky >At 12:40 PM 12/16/97 -0700, you wrote: >> >>Hi Irene, >>Clontech makes FITC labeled as well as Cy3 and biotin labeled annexin V. >>Their number is 800-662 CLON. >> >>-- >>Ronald J.
