Calin I mix the pig blood with 1.5% gelatin (in PBS) equal volumes of blood and gelatin (final gelatin conc. .75%) and sediment for 30 min at room temp. Then I place the supernatant on top of a discontinous gradient of Ficoll-Hypaque (1.119 and 1.077) the PMNs end up at the bottom of the tube with a few contaminating RBCs that you can get rid off with water lysis. I hope this helps Ed ______________________________ Reply Separator _________________________________ Subject: pig PMNs isolation Author: (INTERNET)cta@med.unc.edu at external Date: 12/2/97 6:21 PM Dear Flowers, I am posting this message for a colleague who wants to purify some pig granulocytes. Could you tell me please if there is any particular protocol for this and/or the density of the Fycoll? Thank you, Calin.
