Hi, I am a very new flow operator and would appreciate any comments that might help me resolve my problem. I recently had to analyse a piece of tumour tissue by flow. My results suggest that the sample showed autofluorescence. The original specimen was received in RPMI. From this a cell suspension was prepared in PBS containing BSA and sodium azide. After washing a number of times, the cells were stained, incubated and then analysed. I would appreciate any thoughts about my technique and/or the avoidance of this problem in the future.
