Hi, flowers!

I am a microscopist and now starting the flow. My first experiments did not
work because I lost a majority of the cells during centrifugation. My
questions are:

1. Is it possible to use the indirect reaction or I should bind the FITC
directly to the primery antibody?
2. How can I optimize the centrifugation: time, g, volume, temperature.
3. Is there any good book about the flow for the beginner?
