Hi!
You can get quite near to counting % of S-phase cells  using 
immunohistochemistry (ABC method with microwave) and a monoclonal Ab to 
human Cyclin A (NCL-Cyclin A from Novocastra Lab.,UK).
We have been using it for a while now and the stainings work well even on 
the old archival material.
