Dear Flowers I have been running the TUNEL assay and am concerned that I may have not appreciated the significance of the Ethanol step; I have found I get the better results if I leave the cells in ethanol @ -20oC for some days compared to leaving for say an hour. My question is what would be the minimum and optimal time for this step and can you leave phenotyped cells in ethanol @ -20oC too and for how long? Also what role does it play...? permeabilising the plasma membrane and stabilising the DNA. Many thanks Jane Hughes Immunology Lab Red Cross Children's Hospital Cape Town
