We have the same experience as Tony Bakke. We are very careful when preparing cell suspensions and rarely have any clumping unless the tissue is necrotic. We get the tissue in 30 minutes or less from the O.R. and gently dissociate the cells into RPMI by repeatedly slicing the lymph node with a scalpel. Any non-Hodgkin's lymphoma will pour out into the media with out further manipulations. Hodgkin's disease is another story but then we don't flow those. If you want our procedure for cell suspension preparation let me know. Maryalice Maryalice Stetler-Stevenson Director Flow Cytometry Unit Laboratory of Pathology, NCI, NIH
