O.J.T.jr. I have a Coulter EPICS ELITE ESP with a 544nm Melles Griot 2mW green laser. I have not tried Texas Red with this laser, but I have run rhodamine (TRITC), PE, PI, 7AAD and it works very well. 2mW is sufficient for immunofluorescense, with the right fluorochrome. According to the litterature and other users this laser is also a good exitation source for PE-Cy5, PE-Cy5.5, PE-Cy7, Cy3, Texas Red, LDS751, Po-Pro3, PoPo-3, PyroninY, DiICn(3), DiTBACn(3), SNARF, SNAFL, DiI and PKH26. Another aspect of using 544nm laser is that autofluorescense from mammalian cells is much lower compared to 488nm exitation. This results in higher sensitivity of immunofluorescense measurements. CVs using this laser are as good as 488nm. The cost of this 544 air-cooled HeNe laser is low - another good argument for having this installed. I think Uniphase also have a low-powered 544nm laser.
