I would not expect light scatter pulse width to work. However, a combination of forward scatter, side scatter and pulse rise-time time-of-flight should work. Pulse rise-time is sensitive to the shape, not just the diameter, of cells. If you have access to this measurement on your flow cytometer, use start and stop thresholds of 5% and 35% respectively for greatest sensitivity. -----Original Message----- From: Yashpal Agrawal [SMTP:yagrawal@messi.uku.fi] Sent: Tuesday, March 25, 1997 5:43 PM To: cyto-inbox Subject: flow assay for hereditary spherocytosis I tried to measure pulse width on a FACStar machine for this purpose. The method was not able to differentiate from normal RBC's. Hope this is useful. Yash Pal Agrawal Kuopio, Finland
