For our human progentitor evaluation in blood, marrow, and cord blood we prepare a 'lineage cocktail' of FITC conjugated monoclonals defining mature cells Tcell (CD3), Bcells (CD20) Mono/Myeloid (CD14, CD15, C16) in combination CD34 and other markers of interest for three color analysis; then gating on the Lin- or Lin- CD34+ population. The negative controls we use are are (1) autofluoresence and (2) Lin cocktail + (FL2 or FL3)CD34 + (FL2 or FL3) Isotype. April G.
