Permeabilizing Solution for Intracellular Staining

MAKES 200 mL

    Fill a small beaker with 150 mL of Tris Buffered Saline (TBS).
  1. Add the following while stirring:
    • 0.20 g BSA Sigma A-7030 (0.1% final)
    • 300 uL Triton X-100 Baker X-198-05 (0.15% final)
  2. Adjust pH to 7.4 to 7.5 and qs volume to 200 mL.
  3. Filter into a 1 L orange-top Corning disposable bottle. Store at 4C.

TRIS BUFFERED SALINE (TBS)

MAKES 1 Liter

  1. Fill a 1 L beaker with 750 mL of ddH2O.
  2. Add the following while stirring:
    • 7.88 g Tris HCl Sigma T 3253 (FW=157.6; 50 mM)
    • 8.77 g sodium chloride (FW=58.44; 150 mM)
  3. Adjust pH to 7.4 to 7.5 and qs volume to 1 L.
  4. Filter into a 250 mL orange-top corning disposable bottle. Store at 4C.
