A variety of custom services are available to our clients. These services include peptide synthesis, antisera production, monoclonal antibody development, ascites production, in vitro monoclonal antibody production, monoclonal and polyclonal antibody purification, antibody conjugation and OEM supply. Our professional staff is well trained in all aspects of antibody technology, from production of antisera through immunoassay development. With our extensive experience in purification of both polyclonal and monoclonal antibodies, combined with expertise in conjugation methods employed for labeling antibodies and other ligands with fluorochromes, enzymes and biotin, we can usually provide products to your exact specifications. All custom projects are, of course, held in the strictest confidence.

As a major supplier of antibody-based products, our ability to satisfy our customer's needs and regulatory concerns is important. To meet these concerns, both custom and catalog products are manufactured in compliance with most elements of the FDA guidelines for current Good Manufacturing Practices. For example:

  • SBA manufacturing and control personnel are trained in CGMP regulations which relate to product integrity and quality.
  • The Operations Management Team has well-defined responsibilities and authority for assuring product quality.
  • All materials and components are purchased from qualified vendors, and are subject to formal acceptance procedures.
  • All SBA products, whether catalog, OEM or custom, are manufactured using documented procedures. Prior to release, all products undergo appropriate quality control testing to insure conformance with final product specifications.
  • SBA maintains complete product master records and individual batch history records, thus ensuring traceability for all products from purchased materials through final packaging, and product approval and release.
  • Labeling and packaging materials are strictly controlled for integrity and compliance with specifications.

Antisera Production and Vivarium Services

The SBA vivarium is a United States Department of Agriculture (USDA) licensed facility and follows guidelines set forth by the National Institutes of Health (NIH). Consistent with SBA's Letter of Assurance of Compliance with PHS policy on the Care and Use of Laboratory Animals, which has been submifted to the Office for Protection of Research Risks at the NIH, SBA has established an Institutional Animal Care and Use Committee (IACUC) which reviews each new protocol involving animals prior to project initiation.

Our physical plant includes facilities for ascites production in Balb/c mice, a colony of SPF New Zeland white rabbits, and both indoor and outdoor facilities for maintenance of goats, sheep, donkeys and other host animals employed for antisera production.

Antisera production using antigens supplied by the customer, or produced by SBA, is available on a large or small volume basis. We offer the use of our standard immunization protocol or we can follow a client-supplied immunization procedure and schedule.

NOTE:

SBA cannot accept antigens that are pathogenic, toxic and/or carcinogenic, and certification stating such must accompany all client-supplied antigens.

Prices for this service are divided into three areas: (1) animal cost, (2) per diem cost, and (3) animal disposal. Per diem costs include housing, feeding, yearly vaccinations, veterinary examinations, foot trimming, worming, all antigen injections, test bleeds, production bleeds, and all related materials. All projects have a ninety (90) day minimum time requirement. Please inquire for price quotations.

Peptide Synthesis Antibody Purification

SBA provides services for the small-scale synthesis and purification of peptides to be used as antigens for antisera or monoclonal antibody production. Prices for this service depend upon the length of the peptide to be synthesized, the amino acid sequence, and the degree of purity required for the end product. Purification of antibodies from polyclonal antisera by affinity, ion exchange, gel filtration, hydrophobic interaction, and/or protein A affinity chromatography is available on a small or large volume basis.

Prices for this service are based on a number of factors, including the complexity of the material to be purified, the host species, the quantity of antisera to be processed, and the purity and yield of final product required. Please inquire for price quotations.

Monoclonal Antibody Development

SBAs monoclonal antibody development service consists of five stages: (1) immunization, (2) fusion and selection of hybridomas, (3) screening and expansion, (4) cloning and isotyping, and (5) antibody production / purification. Progress from one stage to the next is always contingent upon client approval. Projects may be cancelled at any time, with payment required only for those stages which have been initiated prior to cancellation. All projects are, of course, held in the strictest confidence. Moreover, any and all monoclonal antibodies produced as a result of this development project shall be the exclusive property of the client.

Ascites Production

SBA maintains its own Balb/c colony for generation of ascites fluid. We approach ascites production from two perspectives:

1. Production of a small pilot batch of ascites for evaluation of titer, antibody concetration, etc. Pilot production allows us to establish realistic manufacturing and quality control specifications, and should be considered by clients who have little or no experience with a particular clone's production capabilities. Once specifications are documented, we can then provide a quotation for bulk production.

2. Bulk production using a specific number of mice to generate a specific volume of ascites fluid is the second option. This option is most appropriate for those clients with established manufacturing and quality control specifications for a hybridoma clone or for those individuals who have used our pilot production service.

In Vitro Production and Purification
of Monoclonal Antibodies

SBA has over seven years experience in "scale-up" production of mouse and rat monoclonal antibodies. Our in monoclonal antibody production service utilizes either a spinner flask for production of small quantities (100-200 mg), or a bioreactor system employing openpore hollow fiber cartridges for large scale production (>500 mg). Production in spinner flasks generally requires 4-6 weeks to achieve targeted production goals. Production in hollow fiber cartridges consists of five stages: (1) mycoplasma testing, (2) assessment of cell viability and antibody production, (3) start-up period of 8-10 weeks to gradually reduce the percentage of fetal bovine serum in the culture media, (4) production phase, and (5) maintenance of production for 6-8 weeks. Monoclonal antibody is then purified from concentrated culture supernatants. Projects may be terminated either at the end of the start-up period or after the production phase, with payment required only for those stages which have been initiated prior to cancellation.

Bulk Production and OEM Packaging Our Best Effort

SBA enjoys several OEM relationships. We can produce large volume lots for enhanced product consistency, and we can package the finished product in either bulk containers or in client-supplied packaging with appropriate labels. Production lots of antibody can be supplied as liquids, frozen liquids, or lyophilized.

Prices for these services are quoted on an individual basis.

While SBA is regarded as a producer of consistently high quality antisera, antibodies, and antibody conjugates, we work on a best effort basis only.
