Antigen distribution:
Cytokeratins are usually expressed in pairs comprising type I and type II cytokeratin. The primary cytokeratins are present in all cell types and are precisely regulated in tissue. Llittle is known about the molecular mechanisms underlying this regulation. In squamous epithelium, as the cells mature and migrate away from the basal layer, the relative contribution of the secondary, differentiated cytokeratins increases at the expense of the primary cytokeratins. Suprabasal layers express specific types of secondary cytokeratins depending upon the site.
Application:
Clone C11 reacts with human cytokeratins 4, 5, 6, 10, 13 and 18. This antibody reacts with a variety of normal reactive and neoplastic epithelia. C11 reacts with simple epithelium and both the basal and suprabasal layers of cornifying and non-cornifying squamous epithelium. Clone C11 may also be applied to staining cultured epithelial cell lines.
It is applicable in immunohistochemistry on frozen sections and methanol/acetone fixed cultured cells. Immunohistochemistry on paraffin, formalin fixed sections requires a protease pretreatment of the deparaffinized sections prior to staining. C11 reacts strongly in immunoblotting.
Specifications:
Monoclonal antibody clone C11 produces mouse IgG1 immunoglobulins which reacts with the human cytokeratins 4, 5, 6, 8, 10, 13 and 18.
