Introduction:
CD54 or intercellular adhesion molecule-1 (ICAM-1) has a wide tissue distribution but is expressed primarily on endothelium in lymphoid tissues and activated lymphocytes (1) . The expression on resting leucocytes is low but is upregulated on activation (2). A soluble form of CD54 is detectable in blood (3). CD54 antigen was initially identified as a widely distributed, cytokine-inducible counter-receptor for lymphocyte-function associated antigen-1 (LFA-1; CD11a/CD18)


Application:
Monoclonal antibodies CD54, clone B-H17 can be applied in biological studies, flow cytometry, or in immuno-histochemistry using cytospots or frozen tissue sections.
Human CD54 is closely related structurally and functionally to CD50 (ICAM-3) and CD102 (ICAM-2) (4). Endothelial CD54 contributes to the extravasation of leucocytes from blood vessels, particularly in areas of inflammation (5). CD54 on antigen-presenting cells contributes to antigen-specific T cell activation, presumably by enhancing interactions between T cells and antigen-presenting cells.
Clone B-H17 monoclonal antibodies have been reported not to inhibit the binding of LFA-1 and do not show reactivity with ICAM-2 and ICAM-3.


Specifications:
Clone B-H17 produces mouse IgG1 antibodies directed against CD54, a 90 - 115 kDa protein. B-H17 was clustered at the Leukocyte Workshop VI (4)


Performance Characteristics:
The CD50 monoclonal antibody, clone B-R1, was analyzed by flow cytometry using blood from a healthy volunteer. Direct staining was performed using 10 ml of FITC conjugated antibody and 100 ml blood sample
