Application of IQ Lyse is recommended using the complete procedure as indicated below (standard method) providing optimal results. Blood samples processed with IQ Lyse will appear in the reagent tube as reddish cell suspension after lysing. The red color does not influence staining results and subsequently flow cytometry analysis.

Standard Incubation Method :

• Add antibody-conjugate to a reagent tube: 10 µl of antibody-conjugate for single reagent, or 20 µl of antibody-conjugate for dual reagent.
• Add 100 µl of EDTA- or Heparin-treated blood and mix well
• Incubate for 15 minutes at room temperature in the dark
• Add 100 µl of IQ Lyse ready to use and mix immediately
• Incubate for 10 minutes at room temperature in the dark
• Add 2 ml of demineralized water and incubate for 10 minutes in the dark
• Centrifuge the labeled cell suspension for 2 minutes at 200xg
• Remove supernatant and resuspend the cells in 200 µl of buffered saline
• Analyze by flow cytometry
