Positive Control Cells for Immunophenotyping

A normal healthy subject was treated with GCSF. A blood sample was taken and the cell count performed. The sample was spiked with serial dilutions of a know concentration of CRISP CD-34 control cells. The samples were then stained with CD-34 and CD-45 moabs. Using a modified ISHAGE acquisition protocol on the flow cytometer, the CRISP CD-34 control cells are detectable to less than 0.1% of the total cell count. Because the CRISP CD-34 control cells are slightly different in size and fluorescence intensity, they can be gated separately and used as an internal control for patient samples.
