Cat. #6576KK: APO-BRDU™ Kit (60 tests). One of the most easily measured features of apoptotic cells is the breakup of genomic DNA by cellular nucleases. The large number of DNA breaks results in an abundance of exposed 3´-hydroxyl termini at DNA ends. The APO-BRDU™ Kit is used to label the 3´-hydroxyl termini with bromolated deoxyuridine triphosphide nucleotides (Br-dUTP). Br-dUTP sites are identified by a fluorescein labeled anti-BRDU antibody. Nonapoptotic cells do not incorporate significant amounts of Br-dUTP as they have few exposed 3´-hydroxyl DNA ends. The kit contains all of the critical reagents required for measuring apoptosis in cells by flow cytometry:

Apo-BRDU™ offers greater sensitivity and more tests than other standard Apoptotic flow cytometry detection systems.

Detection of apoptotic cells by flow cytometry using different methods of DNA strand break labeling. Br-dUTP is more readily incorporated into the genome of apoptotic cells than are the deoxynucleotide triphosphates complexed to larger ligands like fluorescein, biotin, or digoxigenin.(1) This greater incorporation gives rise to a stronger flow cytometry signal when the Br-dUTP sites are identified by a fluorescein labelled anti-BrdU monoclonal antibody. This translates to a greater sensitivity when measuring apoptosis due to better separation of positive and negative cells.
