MoFlo's performance is easy to see. The quality control requirements each unit must achieve before shipment and after installation include:
Sensitivity: <500 MESF*
Purity: >99% (@ 25,000** events/sec)
Recovery: >99% (@ 25,000** events/sec)
Yield: >80% (@ 25,000** events/sec)
CV: <2%
These results are achieved in 1, 2, or 3 laser configurations utilizing non-rectangular gates.
* MLS model. The DTS specification is <800 MESF.
**MLS model. The same results are achieved with the DTS model at 15,000 events/sec.
The reasons for MoFlo's exceptional performance are clear to see in it's list of features, many that are unique in the industry:
Optics
MoFlo is solidly mounted on a sturdy optical table. All components are bolted down, including the lasers, ensuring stability and minimal need for realignment.
Each laser beam has a unique path, independent of other beams. Each laser can be optimally focused without compromise. This means that a 3 laser system is as easy to align as a single beam unit.
All 3 laser beams are spatially separated for minimal crosstalk.
Each laser path is fully enclosed for operator safety.
Fluorescence is focused through separate confocal pinholes reducing noise, crosstalk, and alignment problems.
Two infrared video cameras are utilized for alignment and for monitoring droplet breakoff.
High precision micrometers are used at each alignment point. All adjustments are linear and orthogonal.
Fluidics
The entire fluidics system is designed to operate up to 100 psi with complete safety.
All components are HPLC-grade and color-coded.
Droplet formation rates of up to 200,000/sec. are available.
A biohazard containment system is easily mounted or removed as needed.
Sophisticated pressure regulators with digital displays are used for long-term droplet stability.
The patented laminar flow cell minimizes turbulence and maximizes cell viability at high pressures.
Electronics
With patented parallel signal processing, performance is not affected by using multiple lasers.
There is no instrument limitation to the number of signal channels that can be utilized. Start with a 5, 6 or 7 fl parameter system, and add more as needed (MLS model only).
Unique drop charging circuitry allows 2-way and 4Way sorting (MLS model only).
MixedMode sorting allows you to sort for high purity in one tube, and high recovery in the other tube. This unique feature is vital when the initial number of cells is small.
Instrument controls are available on an easily accessible panel for quick and precise adjustments.
Computer Hardware/Software
A Pentium® computer running Windows® 95/NT is used for instrument control, data acquisition and analysis.
Non-rectangular sort regions are standard and do not affect performance.
Inter- and intra-laser compensation is standard.
CyCLOPS, a native 32-bit software package, maximizes the benefits inherent in Windows 95.
All data are stored uncompensated to allow various compensation schemes to be compared in reanalysis.
Analysis protocols, including gates, annotations, and screen appearance, may be stored for quick recall.
Data files are saved in FCS format.
Accessories
CyCLONE automated high-speed cloning device sorts into 96-well trays, 384-well trays, or any user specified pattern. A 96-well tray can be filled with single cells in <50 seconds.
