Derivatives of the dipeptide lys-ala are preferred substrates of the lysosomal enzyme dipeptidyl peptidase II (DPP II). The dipeptidyl peptidases, a group of enzymes which hydrolyze dipeptides at the N-terminus of proteins and peptides, have a role in processing of proproteins. Some peptides cleaved by dipeptidyl peptidases in nonmammalian systems include yeast pro-a-mating factor, honey bee promelittin, frog caerulein and precursors of cecropins A and B in Hyalophora cecropia. In mammalian systems, DPP II was found in lysosomes of hormone secreting pituitary cells, suggesting that one of its physiological roles may be to digest excess hormones in the secretory granules.
