The recognized epitope is a monomorphic determinant of human HLA class II molecules with estimated molecular weights of 29-34 kDa. These molecules are found on Antigen-Presenting Cells (APC), i.e. B lymphocytes, monocytes, macrophages, and thymic epithelial cells. They are also expressed on activated T cells, but not on granulocytes, platelets, and red blood cells. The I2 antigen is absent from some homozygous typing cells which are DR7+ or DR11+. Resting T cells or endothelial cells that do not normally express these molecules can be induced to express them. These molecules are involved in cellular interactions among immuno-competent cells. I2 antibody was evaluated during the Fifth International Workshop on Human Leukocyte Differentiation Antigens.
