The CDw116 antigen, a 75-85 kDa molecule, is the human Granulocyte-Monocyte Colony-Stimulating Factor (GM-CSF) receptor a chain. It is not expressed by lymphocytes but strongly by monocytes and granulocytes and their precursors. It is also present on endothelial cells, fibroblasts and Langerhans cells. SCO4 and SCO6 antibodies recognize two different epitopes of the GM-CSF receptor a chain. They are both high affinity antibodies which bind to the membrane and to the soluble form of GM-CSFR. They immunoprecipitate the molecule. SCO4 antibody works in Western blot but SCO6 antibody does not. SCO4 is a blocking antibody because it inhibits GM-CSF-driven cell proliferation of TF1, a human GM-CSF-dependent cell line.
