The CD44 family of molecules are transmembraneous proteins with extensive N- and O-linked glycosylation. The standard glycosylated form has a molecular weight of 80-90 kDa. Multiple larger CD44 variant isoforms that arise by alternative splicing of 10 supernumerary genomic exons, have also been described. The CD44 molecules are found on leukocytes (especially on T cells), on red blood cells and weakly on platelets. They are also expressed on bone marrow CD34+ progenitor cells, on brain tissues and epithelial cells. A wide variety of functions have been ascribed to the CD44 molecules including hyaluronate binding, lymphocyte homing to High Endothelial Venules (HEV) and regulation of LFA-3 adhesion. The human blood group antigens Ina/b reside on CD44. J.173 antibody does not inhibit the binding of hyaluronate to its receptor. It can trigger IL-2-dependent proliferation and cytotoxicity of human T cell Clones in vitro. This antibody was evaluated during the Third International Workshop on Human Leukocyte Differentiation Antigens.
