The CD42b antigen (GPIb) is a membrane glycoprotein of 170 kDa, composed by two chains; gpIba and gpIbb, which forms a noncovalent complex with GPIX (CD42a). GPIb a-chain has a molecular weight of 135 kDa. GPIb b-chain has a molecular weight of 25 kDa. This antigen is found on platelets and megakaryocytes. It has also been reported on vascular and tonsillar endothelial cells. GPIbIX complex is known as the receptor for von Willebrand's factor and as von Willebrand's factor-dependent adhesion receptor. The SZ2 monoclonal antibody inhibits the ristocetin-dependent binding of von Willebrand's factor to platelets and ristocetin-induced platelet agglutination. Platelet aggregation induced by type I collagen and Platelet Activating Factor (PAF) is also inhibited. SZ2 antibody immunoprecipitates the components of the gpIb complex. It may also be used in Western blot. SZ2 antibody was evaluated during the Fifth International Workshop on Human Leukocyte Differentiation Antigens.
