The CD34 antigen is a monomeric transmembrane phosphoglycoprotein of about 110 kDa, the extracellular portion of which contains two distinct domains.The membrane proximal domain, of about 110 amino acids, probably adopts a globular conformation. The NH2-terminal domain, of about 140 amino acids, is heavily glycosylated with both N-linked glycans and sialylated O-linked carbohydrates and probably exhibits an extended rod-like structure typical of mucin-like glycoproteins. The CD34 antigen is expressed on hematopoietic progenitor cells of all lineages as well as the most primitive pluripotential stem cells. CD34 antigen expression is highest on the most primitive stem cells and is gradually lost as lineage committed progenitors differentiate. The CD34 antigen is also present on capillary endothelial cells and on bone marrow stromal cells.Variations of glycosylation are thought to occur during normal hematopoiesis depending on lineage committment and the level of cellular maturation. A Number of CD34 antibodies have been described that detect the 3 broad classes of epitopes expressed on some or all of these glycoforms. In order to fully characterize CD34+ cells, four different CD34 monoclonal antibodies are available: Immu133 and Immu409 detect non-overlapping, sialic acid-dependent class I epitopes, QBEnd10, recognizes a Pasteurella glycoprotease-sensitive class II epitope, and Clone 581 recognizes a sialidase / glycoprotease / chymopapain insensitive class III epitope. For quantitation of CD34+ cells in cytokine-mobilized peripheral blood and apheresis products, phycoerythrin conjugates of QBEnd10 or 581 are preferred. Because QBEnd10 detects a denaturation-resistant epitope on the CD34 antigen, it is particularly suited for immunoblotting procedures as well as immunohistochemical analyses on paraffin embedded tissue sections. QBEnd10 antibody was evaluated during the Fourth International Workshop on Human Leukocyte Differentiation Antigens. Antibodies Immu133, Immu409 and 581 were evaluated during the Fifth Workshop.
