The low affinity receptor for aggregated IgG (FcgRIII antigen) exists in two different isoforms; one is a transmembrane form (FcgRIIIA, 50-65 kDa), and the other one is a glycosyl-phosphatidylinositol (GPI) anchored form (FcgRIIIB, 48 kDa). TheFcgRIIIA isoform is expressed on Natural Killer (NK) cells, monocytes and macrophages. The FcgRIIIB isoform is mainly expressed on neutrophils, and is found in two allelic variants, called neutrophil antigen 1 (NA1) and neutrophil antigen 2 (NA2). It exhibits different glycosylation patterns.FcgRIIIA molecule may be non-covalently associated with the g chain of FceRI or with the z chain of CD3. This is the case on NK cells but not on macrophages.The 3G8 antibody belongs to the CD16 family as defined during the Fifth International Workshop on Human Leukocyte Differentiation Antigens. This family of antibodies is capable of binding on FcgRIIIA as well as on FcgRIIIB alleles.
