Hi folks,
While I'm certainly no expert in this area, I feel compelled to
point out that it is the fluorescence _intensity_ of fluorescein
that varies with pH; the peak emission wavelength does NOT change. See
Dr. Haugland's chapter in the latest Methods in Cell Biology manual (part
B), p 658. I had to rebut this point in response to a reviewer's
comments recently, which is the only reason I remember it ;-)

I have seen the same spectral difference with an antibody made here on
campus, but the details of its conjugation were not available. In this
case I suspect Dr. Prussin is on the right track; it's a matter of
FITC =//= fluorescein.
