[Hello Flowers,
Has anyone tried to look at pH measurements on GFP stained cell? We
are concerned about the cross-over of GFP into the Snarf channel -- and
wonder if there are any other pH probes out there that would be at longer
wavelengths. The problem is particularly acute since we are wanting to
do this on an imaging system and compensation may be trickier because
intensities will vary pixel by pixel. Any ideas?]

Dicyanohydroquinone, originally described by Gunther Valet in 1981, is
available as an esterified probe from Boeringer-Mannheim. This is a UV
excited ratiometric pHi probe that should work well with an imaging
system, and avoid fluorescence cross talk with GFP. References can be
found by searching on Gunther, Mike Fox, and in a review that Liz
Musgrove and I published in Cytometry in the mid 1980's.
