TdT Positive AML M3?

Dear flowers

Have you ever experienced TdT positive AML M3?

The patient was in his 30s and the bone marrow
was clouded with typical M3 promyelocytes.
They expressed surface antigen of CD13+and CD33+ without
HLA-DR.
All lymphoid lineage markers were negative.
Our protocol was as follows;
1. Mononuclar cell separation with Ficoll-Hypaque
2. Wash with 1%BSA-PBS once
3. Permeabilization with FACS Brand Lysing Solution
4. Wash with 1%BSA-PBS once
5. Incubation with anti-TdT MAb
6. Wash with 1%BSA-PBS once
7. Data aqusition and analysis with FACSort and HP340
computer system

Autofluorescence without anti-TdT MAb was used as negative
control and MOLT-3 cell line was used as positive control.

About over 90 percent of promyelocytes were nuclear TdT
positive.

What's wrong or was it real?
