We have done several studies using free dye as a standard, The best results we
got were when an intewgrator was used following the preamp. It would put out two
pulses, one for triggering the cytometer and one proportional to the integrated
signal level of the free dye. Such a system requires that you have DC coupled
preamps in your cytometer,

This method, however, is clearly not applicable for day-to-day running and
adjustment of the cytometer. We have had very good results with beads from
Spherotech (1840 Industrial Dr. #270, Libertyville, IL 60048). They make beads
with multiple dyes in them that excite from the UV to the red and are quite
stable over time. We currently use their #NV4C1C18-RM20, which has good signal
and CV in the UV and matches your output spectral reqirements.
