Steve,
You can fix the cells in 4% formaldehyde/PBS and that should keep the
cells fairly stable with respect to protein and DNA. This is the usual protocol
for parafin embedded sections, ploidies are still representitive and p53
levels should not be effected (due to conformational changes, some p53 clones
may not react with formaldehyde fixed cells).
If sterility is required, 20% FCS and 10% DMSO in RPMI is the best
method I know of. Protease activity should be minimal and the DNA stable while
frozen, but protein levels, once thawed, will vary as protein degradation begins.
