It has been a long time since we did any of these staining procedures. A
good reference for procedures is:

Zhong Y-Z dt al Detecting lacZ gene expression in living cells with new
lipophilic, fluorogenic b-galactosidase substrates FASEB 5:
3108-3113,1991.

I believe that you should use the C12FDG rather than the FDG itself. It
is much easier to use and is more sensitive. I would also suggest that you
use the viability stain ( ie FDA ) in a separate tube to compare the
relative fluorescence intensity. We found that FDA was at least 100X more
fluorescent than c12FDG. Good Luck
