In response to TIA's query about range of antibody "concentrations" for
saturation, this depends upon a number of variables including
Number of molecules of target on the cell
Number of cells
Number of epitopes per molecule
Affinity of the antibody
Temperature of the incubation
Length of time of the incubation
Sometimes even wash conditions if a weak antibody.
Amount of nonspecific binding.
A book or article dealing with antibody kinetics would help you
conceptualize these things. The bottom line is you really need to deal
empirically with the issue in your particular system.
