monocellular suspension

In response to Mariangela's question of how to get a monocellular
suspension, it helps to have the cells in Ca and Mg free medium, centrifuge as little as
possible, and pull the cells through a syringe needle (18g). If that
doesn't work, which it usually doesn't well enough for flow, I use a
70-100 micron nylon mesh, filtering immediately prior to analysis. This
works even for the stickiest cell lines for me.
