Hi Birger,

We have used B-B4 on 4 cases involving plasma cell disorders. In
addition we tried known CLL and NHL to check specificity of B cell
stage.

To date no discrepancies have been observed where B-B4 was positive,
and no morphological evidence of plasma cells, or plasmacytoid
lymphocytes.

Plasma cell disorders studied so far show the following:
CD45 CD3 CD19 CD20 CD22 CD56 CD38 B-B4 IgG IgM IgA
I Marrow 21% 8% 0% 77%
I scalp lesion 0% 1% 0% 1% 94% 38% 72% 0% 4%
II LNBX 83% 0% 1% 0% 100 64% + + -
III Marrow 63% 1% 0% 2% 97 40 + +

In these cases cells were labelled with B-B4 and once fixed and
permeableized( Fix and Perm), 2 colour analysis was done for
cytoplasmic expression of heavy and light chains.

The B-B4 antibody was available through Immuno Quality Products.

One of the cases showed a dimmer expression of B-B4. This may be our
in-house problem where using the fix and perm reagent, I lost some of
the antigen.

One of the more recent "markers" proposed for plasma cells in
immunohistochemistry has been the BB4 antibody.
Has anyone information of the corresponding antigen, its
distribution?=
Is the gene cloned and published?
Has anyone been successful using it in FACS-analysis? Would you
detect 100% of myeloma cells???
