Hoechst staining

I do not know whether Hoechst can at all stain a single stranded nucleic
acid molecule. To the best of my recollection Hoechst needs contacts to two
nucleic acid strands while sitting in the minor groove. Data on the mode of
nucleic acid binding have been published during the late 80ties by M.J.
Waring (Cambridge, UK), Stokke and Steen (J. Histochem. Cytochem. (1985) 33,
333), and much earlier by Hilweg and Grop (Exp. Cell Res. (1975) 91, 457).
In addition, Methods in Cell Biology vol. 33 and 41 may contain useful
information.
