Long term use of Fluo-3

Raymond Chang posted a question on long term (90 min) use of Fluo-3. The answer
is (as usual) that it depends to a very large extent on the cells used. In a
lymphoid cell line (CEM-SS) I found that there was good initial labeling but
that by 90 minutes there was overlap between labeled and unlabeled cells. Some
cell types have a Large Anion Transporter that will export all Fluo-3 in ~15
minutes, this transporter can be mostly blocked by probenicid.

I have never attempted to calibrate Fluo-3 levels in terms of uM or nM Ca++, I
seem to remember from old discussions (feel free to help me out here) that the
results were not good enough to be trusted.
