Apoptosis among CD34+ cells

We have been trying to use the TUNEL assay to identify apoptosis in
normal human CD34+ cells double labeled with PE-labeled anti-CD34
antibody (antibody staining before formaldehyde fixation; we have tried
both EtOH and MeOH for the secondary fixation and permeabilization step).
While this is effective for CD34-expressing cell lines, such as TF-1 and
KG1a, in isolated human bone marrow CD34+ cells, the CD34 staining seems
to disappear after the alcohol fixation step- actually down to background
levels.

Anyone have any experience or suggestions?
